Propagation:
The base medium for this cell line is Prigrow II medium available at abm . To make the completed growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 5%, 5 U/ml murine IFN-γ and Penicillin/Streptomycin Solution to a final concentration of 1%. Carbon dioxide (CO2): 5%, Temperature: 33.0°C.
abm does not recommend to use heat-inactivated FBS for cell culture unless specified otherwise.
Quality Control:
1) Western Blot and immunofluorescence were used to detect the presence of SV40 large T antigen; 2) Western Blot was utilized to detect the expression of epithelial markers E-Cadherin and β-catenin; 3) RT-PCR was used to verify the expression of CK8, CK18 and CK19.
