c-Kit or stem cell factor (SCF) receptor is a type III tyrosine kinase receptor that has a major role in the hematopoietic system and has nine key tyrosine residues. Y568 and Y570 residues of the c-Kit receptor are important binding site for signal transduction molecules and contribute to autoinhibition of enzyme activity. c-Kit/Y568F Stable Ba/F3 cells (with GNNK- isoform) has a Y-to-F mutation at position 568, which results in the loss of activation of Src family kinase. The GNNK- isoform is the outcome of the c-Kit gene alternative splicing sites, resulting in different isoforms of the tetrapeptide GNNK, and is the main transcript found in human malignancies.
