c-Kit or stem cell factor (SCF) receptor is a type III tyrosine kinase receptor that has a major role in the hematopoietic system and has nine key tyrosine residues. Y570 residue, alongside with the Y568 residue, is involved in recruiting signaling molecules, and is part of the autoinhibition process of enzyme activity. c-KitY658/Y570F-expressing Ba/F3 cells (with GNNK- isoform) has a Y-to-F mutation at position 568 and 570. The substitution at both binding sites for signal molecules leads to the loss of Src kinase activation, but has c-kit kinase activity with SCF stimulation. The GNNK- isoform is the outcome of the c-Kit gene alternative splicing sites, resulting in different isoforms of the tetrapeptide GNNK, and is the main transcript found in human malignancies.
