Application : The assay measures activity of aggrecanases in the pM-concentration range. It is used to screen and characterize aggrecanase inhibitors.

Description : The assay consists of two moduls, the Aggrecanase Modul and the ELISA Module. A recombinant fragment of human aggrecan inter-globular domain (aggrecan-IGD) is first digested with aggrecanase. Proteolytic cleavage of the substrate releases an aggrecan peptide with the N-terminal sequence ARGSVIL (ARGSVIL-peptide). Subsequently, ARGSVIL-neoepitope containing cleavage products are detected and quantified by ELISA using two monoclonal anti-peptide antibodies.
