To generate the Rieske ISP Stable Knockout Mouse Fibroblast Cell Line, primary lung fibroblast culture was established from a knockin mouse homozygous for the floxed UQCRFS1 (exon 2) gene. UQCRFS1 encodes for the Rieske iron sulfur protein, subunit of complex III. Cells were immortalized by retroviral transduction with E6/E7 type 16 papiloma virus carrying G418 resistance. Clones were obtained from immortalization. One of the clones #8 was used to generate the RIESKE KO cells by transfecting with a plasmid expressing cre recombinase. Clones were obtained after hygromycin selection.
