Applications

The use of MAH00201 in the following tested applications has been tested. The application notes include recommended starting dilutions. Optimal dilutions/concentrations should be determined by the end user. Higher dilution than suggested maybe used in IHC and IF. The product may be used in other not-yet-tested applications.  Refer to Cat # MA00201 for details.

Notes

cELISA: 1/10,000 - 1/100,000 (depending on the amount of coating)

Figure 1:  (Titer of HRP-118-6) The 0.25 pg/ml of SAM coating standard (Cat # ACT00201) was coated on micro-titer wells. The HRP-118-6 was serially diluted and added to the wells. After incubation of about 60 minutes, substrates were added and OD450 was measured.

Figure 2: The standard curve with HRP-118-6 in direct competitive  ELISA using 4-parameter logistic non-linear regression model. The 0.25 pg/ml of SAM coating standard (Cat # ACT00201) was coated on micro-titer wells. Serial dilution of SAM standard (Cat # AST00201) was added along with 1:25,000 diluted HRP-118-6 and was incubated for about 60 minutes. The substrates were then added and OD450 was measured.

Target

Relevance S- Adenosylmethionine is a common co-substrate involved in methyl group transfers. It is made from adenosine triphosphate (ATP) and methionine by methionine adenosyltransferase. Transmethylation, transsulfuration, and aminopropylation are the metabolic pathways that use SAM. Although these anabolic reactions occur throughout the body, most SAM is produced and consumed in the liver.
