The base medium for this cell line is PriGrow IV medium available at abm. to a final concentration of 1% or 35 µg/ml bovine pituitary extract (Hammond), 12.5 ng EGF , 50 µM fresh ascorbic acid, 2 nM estradiol, 1 µg/ml insulin, 2.8 µM hydrocortisone, 0.1 mM ethanolamine, 0.1 mM L-glutamine , 15 nM sodium selenite, 1 ng/ml cholera toxin, and Penicillin/Streptomycin to a final concentration of 1%. For selection use G418 at 100 µg/ml and Zeocin at 20 µg/ml. Adjust media pH to 7.4.
Carbon dioxide (CO₂): 5%, Temperature: 37.0°C.
abm does not recommend to use heat-inactivated FBS for cell culture unless specified otherwise.
