c-Kit or stem cell factor (SCF) receptor is a type III tyrosine kinase receptor that has a major role in the hematopoietic system. The c-Kit wild-type-expressing Ba/F3 cells (with GNNK- isoform) are ideal in studies in relation to c-Kit function, mechanisms, and biological activities that may lead to diseases. The GNNK- isoform is the outcome of the c-Kit gene alternative splicing sites, resulting in different isoforms of the tetrapeptide GNNK, and is the main transcript found in human malignancies. Signaling molecules Cbl, Shc, Gab2, Erk and Akt are more strongly activated by the presence of the GNNK- isoform. These cells will proliferate in the presence of the ligand SCF and activate downstream signal transduction. Rapid degradation of the SCF is observed in cells expressing the GNNK-, as the Src inhibitor SU6656 (SUGEN) exerts stronger effects when compared with GNNK+ isoform cells.
