Product Overview
Gellan Gum is an anionic polysaccharide used as a gelling agent and as an alternative to agar in plant tissue culture and selected microbiological media. Its repeating tetrasaccharide structure contains two D-glucose residues, one L-rhamnose residue and one D-glucuronic acid residue.
In plant tissue culture, Gellan Gum can produce a firm, relatively transparent gel that makes it easier to observe plant tissues and root development. Depending on the culture medium and protocol, it may be used at a lower concentration than agar.

Gellan Gum may be used at approximately 0.2–0.4% (w/v), equivalent to 2–4 g/L, to solidify plant tissue culture media. The optimal concentration depends on the medium and application.
Example: Preparation of 1 L Plant Tissue Culture Medium
The following is a general preparation outline. Follow the validated protocol for your basal medium and culture system.
- Add approximately 800 mL of distilled water to a suitable beaker.
- Add 4.54 g of MS basal medium (or an appropriate alternative) and 30 g of sugar.
- Add PPM™ if required by the selected protocol; a typical example is 1–2 mL/L.
- Bring the solution to a final volume of 1,000 mL.
- Adjust the pH to approximately 5.4–5.8.
- Add 3–4 g/L of Gellan Gum while stirring.
- Autoclave using a validated cycle. A commonly used cycle is 121°C, 15 psi for 20 minutes, subject to the medium and vessel load.
- Allow the medium to cool and solidify before use.
Note: Medium composition, pH, gelling-agent concentration and sterilization conditions should be optimized for the plant species and laboratory protocol.
