Description
Protein activity and functional studies often require purified target proteins. Many recombinant proteins are expressed as fusion proteins containing polyhistidine purification tags, commonly known as His-tags.
His-tags are widely used because of their small size, relatively mild purification conditions, and compatibility with metal-affinity chromatography. Ni-IDA Spin Columns are designed for rapid, small-scale purification of polyhistidine-tagged recombinant proteins.
Each column contains 100 µL of high-performance Hi-Bind™ Ni QR Agarose Beads (Cat. #6562), enabling the capture and purification of up to 2.8 mg of polyhistidine-tagged protein per column under suitable conditions.
The compact bead volume supports the processing of different sample amounts. Under appropriate conditions, the tagged target protein binds to the nickel-based affinity resin while many untagged contaminants are removed during washing.
The columns are compatible with centrifugation-based processing, helping reduce purification time compared with some conventional workflows. Protein recovery and purity depend on the protein's expression level, structure, solubility, and the purification conditions used.
Key Features
- Designed for purification of polyhistidine-tagged recombinant proteins.
- Nickel-based immobilized metal affinity chromatography (IMAC).
- 100 µL of Hi-Bind™ Ni QR Agarose Beads per column.
- Purification capacity of up to 2.8 mg of His-tagged protein per column, depending on the sample and conditions.
- Centrifugation-based format for rapid small-scale purification.
- Pack contains 25 spin columns.
Principle of Ni-IDA Affinity Purification
Ni-IDA chromatography uses nickel ions coordinated by an immobilized iminodiacetic acid (IDA) ligand. Histidine residues in a polyhistidine tag interact with the immobilized nickel, allowing the tagged protein to be retained on the resin.
Unbound sample components can be removed during washing. The target protein is then recovered using an appropriate elution condition, commonly involving imidazole or another compatible method specified by the manufacturer's protocol.
Applications
- Purification of His-tagged recombinant proteins.
- Small-scale protein expression screening.
- Preparation of proteins for biochemical and functional studies.
- Purification of recombinant proteins before downstream analysis.
- Research workflows involving affinity chromatography.
