Monocyte Chemotactic Protein-1 (MCP-1), also known as Chemokine (C-C motif) ligand 2 (CCL2), is a member of the beta (CC) subfamily of chemokines. MCP-1 is characterized as a monocyte-specific chemoattractant. Because of its target cell specificity, MCP-1 is postulated to play a pathogenic role in a variety of diseases characterized by mononuclear cell infiltration, including atherosclerosis, rheumatoid arthritis and allergic responses. In basophils, MCP-1 is highly effective as a stimulus of histamine release but has only weak chemotactic activity. Additionally, it has been shown to chemoattract CD4+ and CD8+ T lymphocytes.
The Mouse MCP-1 ELISA is an enzyme-linked immunosorbent assay for the quantitative detection of mouse MCP-1 in different tissue plasma, serum, and other biological samples. The plate has been coated with murine MCP-1 monoclonal antibody. Any MCP-1 present in the sample or standard will be captured. A biotin-conjugated anti-murine MCP-1 antibody is added followed by streptavidin-horseradish peroxidase (HRP) and an accompanying substrate. The reaction exhibits a blue color in direct proportion to the amount of MCP-1 in the initial sample. The reaction is terminated by the addition of a stop solution, and absorbance is measured at 450 nm. The concentration of MCP-1 in the samples is calculated opposite to the standard curve.
