This rat cell line expresses a chimeric molecule containing a Δ E-like fragment of Notch1 fused to a vesicular stomatitis virus epitope, the Cre recombinase, and a modified hormone binding domain of the estrogen receptor. The delivery of Cre recombinase depends on any event, including γ-secretase processing, that releases the intracellular domain of Notch1 from the membrane. Nuclear activity of the Cre-ER-T2 chimera depends on the presence of tamoxifen. Reporter construct for the recombinase activity consists of an antibiotic resistance gene (hygromycin). This cell line would be well-suited for research regarding the functions of the Notch receptor, γ-secretase enzyme activity, γ-secretase multiprotein complex, and their involvement in cellular signalling.
